Martin learns that Lucy's husband has been missing for a long time and he befriends her children, Sass and Bike. When Martin goes to the village, he has a hostile reception from the locals. Along the days, Martin spends his days in the Tasmanian wilderness chasing the Tiger and becomes closer and closer to the Armstrong family. - Mercenary Martin David (Willem Dafoe) is hired by military biotech company Red Leaf to go to Tasmania and gather samples of the Tasmanian tiger, with further instructions to kill all remaining tigers to ensure no competing organization will get their DNA. The deal is set up by a middleman (Jacek Koman). The Tasmanian tiger is believed to be extinct and it the rarest, most exclusive creature on the planet. Red Leaf has had 2 confirmed sightings of the animal in Tasmania. The news will spread quickly, so Martin has only a few weeks to complete his mission. Martin is a hyper organized and disciplined person who can hold his breath for extended periods of time.
Martin flies into Hobart to start his mission. Posing as a university biologist, Martin lodges in the home of the Armstrong family: Lucy (Frances O'Connor) and her two young children Katie (Morgana Davies) and Jamie (Finn Woodlock). The house does not have electricity most of the time and relies on a generator that doesn't work. The taps give dirty water and generally the house is in a bad condition. Martin tries to rent a room at the local tavern, but they refuse, and he has no option but to stay with Lucy.
Lucy is perpetually benumbed from prescribed medication, taken after the disappearance of her environmentalist husband, Jarrah Armstrong (Marc Watson-Paul). Lucy spends most of her time sleeping due to the medications. Speculation surrounds Jarrah's disappearance, particularly with regards to a longstanding conflict between the local loggers who are in desperate need of jobs, and the 'greenies', a group of environmentalists who have set up roadblocks to the forest to prevent its deforestation. The town is hostile to Martin as they believe that he is another environmentalist like Jarrah.
Martin goes about repairing the house, including the bathtub and the generator. Martin goes into the bush for twelve days at a time, setting up various steel traps and makeshift snares, while waiting patiently to see if a tiger will surface. Martin carefully marks the position of his traps on his map of the jungle. Martin is adept at living off the land and is a good hunter. Martin hears gunshots in the jungle, which indicates that there is a rival hunter, or it could be the locals who want him to go back. He also returns to the edge of the jungle to find his car vandalized.
During his short stays at the Armstrong's to resupply, Martin slowly befriends the children, and discovers that Lucy's medication is delivered to her by Jack Mindy (Sam Neill), who has been unofficially looking in on the family. Martin confiscates Lucy's medication, and bathes her while she is unconscious, after realizing the detrimental effects of her dependency.
During one return from the bush, Martin finds Lucy has recovered from the symptoms of her addiction. Jamie provides Martin with a clue as to the tiger's whereabouts: a drawing of the tiger near trees and small bodies of water. From the drawing, Martin is able to deduce the tiger's location on his map. He also finds other traps in the jungle, which confirms his suspicion that another hunter is operating, and may have the same objective as Martin. He finds an automated camera and takes away its memory card. Martin returns home and checks the memory card and finds nothing useful on it. On his next trip out, Martin stumbles across Jarrah's skeletal remains and discovers that he had been shot through the head. Martin gives him a proper burial but does not reveal his findings to the Armstrong family. On his return to the Armstrong house, Lucy informs him that Red Leaf had initially contracted Jarrah to locate the tiger, a pursuit he eventually abandoned in favor of taking up an environmental cause to protect wildlife and that Red Leaf wanted Jarrah to find the tiger because they believed that it had a paralyzing venom in its bite.
While hiking to check his traps, Martin is ambushed by a rival Red Leaf operative (Callan Mulvey) sent to replace him. (The Rival operative had entered Lucy home to check the maps that Martin left there, which revealed his location. While there, it is implied that the rival operative set fire to the home).
The man binds Martin's hands and instructs Martin to lead him to the tiger's cave, but Martin instead leads the operative past one of his steel traps. The operative steps on the trap, and its metal teeth bind his leg. The operative drops both rifles. Martin frees his hands, picks up one of the rifles, and kills the operative just as the operative frees himself from the trap and lunges for the other rifle.
Martin returns to the Armstrong residence to find it burnt down. Confronting Mindy, he learns that Lucy and Katie had perished in the fire that broke out by accident, but Jamie survived and was taken by the authorities. Martin sets out into the bush once more to find the Tasmanian tiger and put an end to Red Leaf's pursuit. He finally finds the creature and reluctantly shoots it, then proceeds to cremate it in order to remove all traces of its existence.
Martin returns to town and calls Red Leaf, informing them that what they are looking for is "gone forever". He then goes to a school where Jamie sits alone on a bench. When Jamie sees Martin, he runs excitedly toward him and the two embrace.
